Installing 16 x 16" wall ceramic tile?

Never installed the big boys before. How long do I have to hold those things against the wall before I can move on to the next tile (where they don't fall down)? Also, do you need quite a bit more thinset behind each tile than you would with, say, 6x6 inch tile? Tiling against well-secured Hardie Backer Board
